The sophisticated and multi-disciplinary approaches encompass immunology, virology and molecular biology.
The main immunological focus is on T lymphocyte biology. Lymphocyte biology work will use high dimensional flow cytometry to study T and B cells in the context of immune deficiency, both Primary immunodeficiencies and acquired immunodeficiency due to HIV infection, as well as highly immunosuppressed patients.
Assay development work will include development of novel diagnostic tests of the immune system including deep immune phenotyping using multiparameter flow cytometry, cytokine testing, cytokine autoantibody testing, immune signalling pathway testing and inflammasome pathway testing.
Studies of HIV include investigation of memory CD4 T cells as a therapy-resistant reservoir of HIV that can reignite the infection if therapy is interrupted. CD4 T cells are being isolated and cultured to show viral replication in vivo and in vitro, using a newly developed and patented PCR assay technique.
Another important aspect of CD4 T cell biology is antigen-specific memory cells, which we study using another patented flow cytometry method. This has opened up opportunities in the study of other viral infections, including the possible role of EBV in multiple sclerosis, as well as study of immune responses in TB infection.
